실험동물과 실험용 동물(experimental animals)이라는 용어는 비슷한 의미로 혼용되어 사용되고 있으나, 엄밀한 의미에서는 확실히 구별되어야 한다. 실험동물이란 마우스(실험용 생쥐), 랫트(실험용 집쥐), 마모트, 터쥐류, 실험용 토끼 및 특정 종류의 개나 고양이 등을 말한다. 그러나 동물실험에 사용되는 동물은 이보다 훨씬 많아서 가축(소·말·면양·산양·돼지·닭·개·고양이 등), 야생동물(원숭이류·설치류·조류·파충류·양서류·어류·곤충류) 등도 포함되기 때문에 이러한 모든 것들을 총칭해서 실험용 동물이라고 부르는 것이다. 실험용 동물은 원생동물로부터 포유동물까지 모두 포함되나, 실험동물은 이 중에서 가축이나 야생동물과는 달리 동물실험의 정밀도와 재현성을 높이기 위하여 연구 목적에 맞도록 육성·번식·생산된 동물이다. 실험동물은 실험 목적에 따라 선택되는데, 일반적으로 실험실 내에서 계대배양(繼代培養)이 가능해야 하며, 사육이 간편하고 온순하여 다루기 쉽고, 번식력이 강하고 건강해야 한다.

이러한 실험동물을 이용하여 처치를 하고, 그 결과로 나타나는 동물의 생체반응을 관찰·측정·해석하는 것을 동물실험(animal experiment)이라 한다. 한편 생물검정(biological assay/bioassay)이란 과학의 발달에 의해 새로 만들어지는 신물질이나 자연에서 추출되는 수많은 약물, 화학물질, 생물학적 제재 등의 효력의 단위, 또는 역가(potency)에 관해서 실제로 사용하기 전에 동물을 사용하여 정성·정량적으로 검정하는 것을 말한다. 이와 같은 동물실험은 실험동물의 유전적 요인, 성별, 나이, 기타 생리요인, 질병요인(건강상태), 영양요인 등에 의해 크게 영향을 받게 되므로 이들을 고려하여 실험에 사용해야 한다. 실험동물을 유전적으로 동계교배계(inbred strain)·돌연변이계(mutant strain)·폐쇄군(closed colony)·교잡군·잡종으로 나눌 수 있는데, 유전적 요인에 대한 고려로서 실험 목적에 따라 이들을 선택하여 사용한다. 동계교배계는 형매교배나 친자교배를 20세대 이상 계속한 동물이다. 이와 같은 교배를 20세대 이상 하면 동계교배계수(F)는 98.6％, 혈연계수(R)는 99.6％가 되므로 수 세대 후에 같은 실험을 하더라도 같은 성격이 기대된다. 동계교배계 동물을 유지하는 방법은 형매교배나 친자교배를 계속 유지하는 것이다. 현재 국제적으로 등록되어 있는 동계교배는 마우스가 약 300계통, 랫트가 약 110계통으로 여러 가지 특성이 조사되어 있다. 폐쇄군은 5년 이상 외부의 이입없이 일정한 집단으로 번식시키고 있는 동물로서 마우스는 ICR·ddY, 랫트는 SD·위스터(Wistar) 등이 있으며, 동계교배계와 비교하여 번식력이 좋아 다루기 용이하다. 교잡군은 동계교배계 간의 교배에 의한 1세대(F1)를 주로 사용한다.

 

 

실험동물은 살아 있는 시약이라고 불릴 정도로 실험의 성패를 결정하는 중요한 요인이다. 동물실험의 결과에 신뢰성과 재현성을 부여하기 위해서는 유전적 조절, 질병 조절, 환경 조절이 되어 있는 실험동물 계통을 사용해야 한다. 동물실험에는 양질의 동물을 사용하는 것이 바람직하나, 목적에 따라 질에 관한 선택을 해야 한다. 유전적 조절은 실험동물을 유전적 균일성을 갖게 하기 위해, 선발 및 도태를 통하여 육성하거나 근친교배에 의하여 육성한다. 또한 엄격한 관리하에서 계통을 유지하며 계통간의 교잡, 돌연변이, 관리상의 실수에 의한 오염을 막기 위해 정기적인 유전적 분석을 해야 한다. 실험동물이 질병을 앓고 있다면 이들을 사용한 동물실험의 결과는 매우 달라지기 때문에 질병 조절이 된 건강한 실험동물을 사용해야 한다. 병원미생물이나 기생충 등의 질병을 일으키는 원인체의 존재여부에 의해서 실험동물을 분류할 수 있는데, 모든 미생물이나 기생충이 없는 무균동물(germ free animal), 동물실험의 결과에 영향을 미치는 미생물이나 기생충이 없는 SPF(specific pathogen free) 동물, 그리고 미생물 조절을 전혀 받지 않은 일반동물(conventional animal) 등으로 나눌 수 있다. 무균동물과 SPF 동물의 사육환경은 청정공기를 넣어주며, 사용하는 기구 및 기재는 모두 멸균 소독된 것을 사용하고, 사육실 내 사람의 출입을 엄중히 제한시키며 의복도 멸균된 것만 사용한다. 실험동물은 정기적으로 미생물 검사를 하여 오염을 막아야 한다. 환경조절은 실험동물 사육의 온도·습도·영양상태·조명·소음 등의 요인들을 일정하게 조절하는 것을 의미한다. 한편 실험목적의 다양화에 의해 실험동물도 다양화되어 세포공학 및 배(胚) 조작을 응용한 유전자도입 마우스(transgenic mouse), 키메라 동물, 핵이식(nuclear transplanation)에 의한 새로운 실험동물 등이 만들어지기도 했다. 또한 인간이 직면하고 있는 암, 유전성 질환, 만성 성인병 등의 연구 및 치료를 목적으로 하는 실험에 사용되는 질환 모델 동물들도 다양하게 개발되었다.

Hybrids are very valuable for research. The features and uses are as follows:
Genetic and phenotypic uniformity.
Increased disease resistance, better survival under stress, greater natural longevity,larger litters.
Accept transplants of tissue(tumor, skin, ovary) from mice of either parental strain.
Useful in screening tests for anti-cancer agents.
B6C3F1 hybrid is useful in long-term studies and carcinogenecity studies.
