In mouse experiments involving several tumor models, the researchers report that certain LNP formulations promoted the spread of tumor cells, including increased metastatic tumor burden in the lungs; importantly, the effect was also observed with empty LNPs containing no mRNA, suggesting that the nanoparticle formulation itself contributed to the effect. The researchers propose that injected LNPs caused local tissue and mitochondrial damage, releasing mitochondrial DNA (mtDNA) into circulation; this activated neutrophils and triggered NETosis, in which neutrophils release web-like structures called neutrophil extracellular traps (NETs). According to the study, this inflammatory environment made it easier for circulating cancer cells to establish metastatic tumors, and interfering with neutrophils or NET formation reduced the effect.
